Annotation description: | Experiment explanation: The authors systematically characterized the molecular and morphological nature of the docked and primed SV states with nanometer accuracy by the used of high-pressure freezing (HPF), freeze substitution (FS), and electron tomography (ET) of mouse mutants synapses. Taking in account as analysis parameters: SV Diameter and Number of SVs within a distance of: 100, 40, 0-5, 0-2nm of AZ. Protein Function: The analysis of Syb- 2 KO terminals revealed a severe reduction in the number of docked SVs and a prominent accumulation of SVs close (4-8 nm) to the AZ (Figure 4).However, Syb-2 KO synapses showed no changes in the number of SVs within 40 or 100 nm of the AZ. Based on these results, the authors concluded that Syb-2 is required for SV Docking. Protein Location: Confocal microscopy of WT slices showed a specific punctate labeling of Syb-2, which highly colocalized with VGLUT1 but was absent in Syb-2 KO cultures (data not shown). |
