Annotation description: | RNF-10 is a protein with synaptic and nuclear localization that contains two NLS within its sequence, suggesting its role as a synaptonuclear messenger. Treatment with Bicuculine to increase network activity induced a reduction of RNF-10 immunoreactivity in synapses and a simultaneous enhancement in the nucleus. This effect is nicely monitored by using an photo convertible RNF-10, which is photo converted in distal dendrites upon application of bicuculine and whose fluorescence can be observe in the nucleus over time. Translocation of RNF10 is dependent on activation of synaptic NMDA receptors. Interestingly, RNF10 can interact with importins alpha, and bicuculin treatment enhanced the interaction between RNF10 and importin alpha1, suggesting an importin-dependent nuclear translocation of RNF10. Photoconversion experiments performed after induction of LTP using field stimulation reveals that RNF10 accumulated in the nucleus after long distance protein transport. This was corroborated using a different approach in which levels of RNF10 in the nuclear fraction were assessed after induction of cLTP in primary hippocampal cultures, which revealed an enrichment of RNF10. Finally, downregulation of RNF10 levels by means of a shRNA revealed that lacking of RNF10 prevented expression of target genes such as p21waf/cip1 and Ophn1. |
