Annotation description: | The authors use the drosophila NMJ to study genes involved in the stable formation of synapses. Using an RNAi screen they identify CK2b as essential for synapse formation. -Knockdown of CK2b let to the formation of postsynaptic structures (marked with dGluRIII) that are not opposed to presynaptic terminals (marked with Brp). A similar phenotype was observed in an CK2b hypomorph (CK2b-P1) and an near-complete knockout (CK2b-P1/CK2b 26-2). (Figure 1) -Only presynaptic CK2b expression is required, since knockdown in the neuron, but not in the muscle, induces the phenotype (figure 2). Additionally, the phenotype can be rescued by presynaptic, but not postsynaptic re-expression of CK2 (Figure 3). -The knockdown efficacy is demonstrated via western blot (figure 2K). Fly CK2b has 88% homology with mouse CK2b (main text) 24/11/2017 Pim - I have selected the 'regulation of ...' variant since CK2 is a signaling protein. |
