Annotation description: | An knockdown for liprin a2 (a2 KD) is using in rat hippocampal cultures to study its role in synapse morphology using electron microscopy. a2 KD induced a range of relatively mild effects on synapse morphology, including a reduction of the number of docked vesicles, active zone length and the density of vesicles. The total size of the vesicle cluster, however, was slightly increased. This suggests that liprin a2 plays a role in the assembly and/or maintenance of the presynapse, including the active zone. This is somewhat consistent with the phenotype of Liprin mutants in c.elegans (Zhen & Yin) and drosophila (Kaufman et.al.), although in these studies a broadening of the active zone was observed, with no effect on docking. The knockdown is validated in several ways. In western blot, the shRNA only affects 2a levels, without affecting other liprins (Figure 2 A-F). The shRNA also appears the liprin 2a signal at synapses in confocal microscopy (Figure 2D). Overexpression of liprin 2a can rescue the evoked excitatory current to near wild type levels (figure S1). An independent shRNA shows a similar phenotype (figure s1). |
