Annotation description: | The paper studies the distribution of NKAα1 and NKAα3 in the rat spiral ganglion and in the organ of Corti Figure 5 demonstrates membrane staining for NKAα3 directly surrounding cytoplasmatic synpasin staining at the bouton-shaped terminals of the medial olivocochlear efferents that contact the outer hair cells (Fig. 5D-I). The expression of synapsin is known to label these presynapses. Double labeling of organs of Corti with a monoclonal anti-NKAα3 (Fig. 5A) and a polyclonal anti-prestin (Fig. 5B), a protein enriched in the cell membrane of the postsynaptic OHCs, shows the NKAα3-positive fibers traveling through the tunnel of Corti and contacting the OHCs with their bouton-shaped terminals (Fig. 5C). There is no apparent postsynaptic labelling. Additional literature: | Figure 7 presents an immunohistochemical analysis where the presynaptic terminal of the calyx of Held synapse was filled with Alexa 488 and following fixation, the slice was stained for the presynaptic marker Vglut1 and for the the Na+/K+-ATPase alpha3 using an isoform-specific antibodies. Strong staining was observed in the presynaptic compartment, but not in the postsynaptic compartment. However, it is hard to determine the sub cellular compartment in which the staining is found, as the signal is very diffused. In the text, the authors report a negative staining for the Na+/K+-ATPase alpha1 isoform. @ PMID:17220883 |
