Annotation description: | Spontaneous fusion frequency is increased in slices (Fig 6D) while the amplitude of the spontaneous responses (Fig 6A) and number of synapses (Fig 6B) remain the same in the pfn2 null mutant. Increasing the extracellular calcium also produced a bigger EPSC amplitude in pfn2 KO mice (Fig6E). These data show that the vesicular release probability is increased in absence of Profilin2. This could be related to altered presynaptic actin dynamics since depolarisation-induced actin polymerisation is synaptosomes from pfn2 null mice was impaired (Fig 7C) and no direct effects on proteins level of the release machinary itself were detected (Fig 7B). |
