Annotation description: | The authors generate a syndapin 1 KO mouse. In figure 3 they use FM1-43 uptake assays to measure endocytosis in cultured hippocampal neurons. At low frequency stimulation there was a very small decrease in FM1-43 dye uptake but at higher frequency stimulation there was larger relative decrease in SV endocytosis when comparing KO to wt. In figure 4 they use EM in the mouse retina. This synapse fires extensively in the dark. Shifting to light causes a strong compensatory endocytosis. The synapses from KO mice showed numerous irregularities when shifting to light including accumulation of clathrin-coated pits at the plasma membrane and alterations in SV size. In figure 7, the authors use whole-cell recordings from CA1 pyramidal neurons of syndapin I KO mice and observe reduced frequencies of miniature excitatory and inhibitory potentials consistent with a defect in synaptic vesicle recycling in syndapin I KO neurons. 19/6/2017 Pim - Literal: "Detailed molecular analyses demonstrate that syndapin I plays an important role in the recruitment of all dynamin isoforms, central players in vesicle fission reactions, to the membrane." Since syndapin seems involved in the recruitment of dynamin I do no classify this protein as a "regulator of..." SVE. |
