Annotation description:Study in the calyx of Held synapse to determine the effects of NCS-1 infusion into the presynaptic compartment.

Figure 1a demonstrates that the infusion of NCS-1 leads to a transient (circa 5 minute) facilitation of the presynaptic Ca2+ current (measured very precisely in this synapse).

Figure 1b demonstrates a negative shift in the current-voltage (I-V) curve of Ca2+ channels, suggesting a direct effect of the NCS-1 overexpression on Ca2+ channel properties. The inclusion of BAPTA together with NCS-1 abolished the effect on the I-V curve, suggesting that the effect of NCS-1 on the Ca2+ channels is Ca2+-dependent. Specifically, the 10 to 90% rise time of the Ca2+ current is faster is calyces where NCS-1 is infused, indicating a direct effect on channel properties.

Figure 2 demonstrates that the Ca2+ current facilitation caused by the perfusion of NCS-1 occludes the Ca2+ current facilitation that is usually observed during paired-pulse stimulation.

A C-terminal peptide of NCS-1 that lacks the Ca-binding domain does not show the effect mentioned above, but blocks the effect of infused full length NCS-1 (Fig.4). This suggests that the Ca-effects are not mediated by a Ca-binding/buffering function of the protein, and are likely the result of direct interaction with the target protein (VGCC?). The SyNGO term 'calcium ion binding involved in regulation of presynaptic cytosolic calcium levels (GO:0099534)' is not relevant therefore for the NCS-1 protein.
