Annotation description: | Clathrin-coated vesicles (CCVs), which by nature are coated with the annotated protein, clathrin heavy chain (CHC), are purified from pre-synaptic nerve terminals. The authors demonstrate in figure 2 that the CCVs are from the pre-synaptic compartment because they require osmotic shock to be released (because they are trapped in intact synaptosomes) whereas CCVs from other parts of the cell, including the post-synapse, would not require this treatment. In figure 4 the authors demonstrate by coomassie staining and western blot that the CCVs are highly enriched in CHC and multiple synaptic vesicle (SV) proteins, clearly indicating that CCVs are involved in SV recycling. Figure 7 uses immuno-electron microscopy to demonstrate that CCVs isolated from the pre-synaptic compartment and whole brain show very similar levels of staining for the SV protein synaptophysin, indicating that the major localization of CHC in brain is on CCVs that recycle SVs. 22/5/2017 Pim - I agree with the expert.
