Annotation description:Pre-embedding immuno-EM indicates immunoreactivity for Complexin-2 in axon terminals (vesicles, plasma membrane) and it filled the axoplasm in a varying degree in different synapse types and brain regions. EM localization indicates that a larger proportion of the protein may be associated to membranes (in comparison to the subcellular fractionation, where most of the synaphins are in the soluble, cytosolic fraction - Referenced to SynGO annotation #577 ).

monoclonal antibodies (mAbs) specific to synaphin 1 (12C5) were generated in the study. No KO control.

Rationale:
Complexin-2 (Synaphin; 19a) in the brain is more concentrated in the soluble fraction (lane 2) than in the insoluble fraction (lane 3) (Fig.4A) and is therefore mainly located in the cytoplasm.

"Complexin was scarce in the LP2 fraction (that is rich in synaptic vesicles), indicating that very little, if any, of the protein is associated with synaptic vesicles."(Fig. 5)

Complexin in the insoluble fraction is more likely to be on the plasma membranes than on SVs.
