Annotation description: | Complexin-1 is present in the presynapse (co-localizing with the synaptic vesicle marker Vglut1) in the Calyx of Held terminal of the MNTB in the brainstem (Fig.1C). Complexin-2 expression is downregulated in Calyx of Held terminals during development (Fig.11). At P21, Vglut-1 immunoreactivity does not colocalize with with Complexin-immunoreactivity in Complexin-1 KO mice (Fig.11C). Therefore, the immunoreactivity in wild-type Calyx of Held terminals at P21 must originate from the presence of Complexin-1 in the presynapse (Fig.1C). Complexin-1 immunreactivity is present in the somata of the MNTB principal neurons indicating also a postsynaptic localization. IHC was performed using an antibody (SySy, rabbit anti-Cplx-1/2) that detects Complexin-1 and -2. Complexin-1 KO mice exhibit a loss of immunoreactivity in the somata of the MNTB principal neurons (Fig 11C) indicating the specificity of the antibody. |
