Annotation description: | Postsynaptic knock-down (KD) of Complexin-1 and -2 (shRNA stereotactic in vivo injection into CA1 region of the hippocampus) blocks LTP (Fig. 1 F), indicating that Complexin plays a role in postsynaptic membrane fusion events underlying the exocytosis of AMPA receptors during LTP. NMDA-Receptor mediated LTD is unaffected by postsynaptic Complexin KD (Fig. 1 H). Block of LTP can be rescued by expression of a shRNA-resitant full-length Complexin-1 variant (Fig. 1 G). Basal synaptic transmission is not affected by postsynaptic Complexin KD in slices (Fig.2) and cultures (Fig 4B), indicating that Complexin only regulates AMPA-Receptor exocytosis. |
