| Annotation description: | CAPS associates with synaptic DCVs. Fig.4 Immunogold labelling of lysed synapsosomes.The CAPS polyclonal antibodies used were generated either against a baculovirus-encoded rat CAPS protein (Ann et al. 1997) or against E. coli-expressed Drosophila CAPS fusion proteins (B. Berwin, unpublished data) and were affinity purified by the method of Olmsted 1981. Control studies conducted by immunofluorescence demonstrated that immunostaining was reduced to background in the presence of an E. coli-produced recombinant CAPS protein (J. Wassenberg, personal communication). In the immunogold procedure, gold-conjugated secondary reagents were used at a dilution where no labeling was observed in the absence of primary antibody. Fig.5 DCV and synaptic vesicle (SV) fractions from rat brain homogenates were prepared and tested by Western Blot using CAPS polyclonal antibodies (no KO control). Fig 5A demonstrates an enrichment of CAPS in the LDCV pool in comparison to the SV pool. Fig 5B and C: CAPS cosediments with DCVs, but not SVs using a sucrose gradient. Fig. 5D: Treatment of DCV fraction with proteases cleaves VAMP2 and CAPS, but not a DCV content marker, indicating that CAPS is an extrinsic component of neuronal DCVs. 31/5/52017 Pim - I agree with the plasma membrane localization not being annotated becase these experiments do not distinguis pre- form post-synaptic localization. |
