Annotation description:Figure 2a, 3e: immunofluorescence imaging of hippocampal neurons stained for endogenous ELMO1 and PSD95 (DLG4) showed partial co-localization. Overexpressed GFP-ELMO1 localized to dendrites and spines apposed to presynaptic terminals labeled by anti-SV2.

Figure 2c-d, 3: confocal imaging showed that ELMO1 shRNA knockdown reduced dendritic spine density in hippocampal neurons and overexpression of the ELMO1 C-terminus promoted spine morphogenesis.

Figure 4: ELMO and Dock180 (DOCK1) form a complex in hippocampal neurons and function through activating Rac1.

Figure 5-7: RhoG is required for spine formation, its activation leads to increased spine head size and functions through the ELMO1/Dock180/Rac pathway.
