Annotation description: | Figure 1a, S1: generation of a polyclonal pan-DCLK-antibody against the DCLK1 DCX domain. Targets a region with high homology between DCLK1 and DCLK2, has higher affinity for GFP-DCLK1 than GFP-DCLK2, and was validated with DCLK1-/- and DCLK2-/- knockout mouse controls. Figure 1i-j: immunofluorescence staining for endogenous PSD95 (DLG4) and overexpressed GFP-DCLK1 in cultured hippocampal neurons showed DCLK1 localization to dendrites and dendritic spines. Figure 1k: DCLK was enriched in SV, synaptic plasma membrane and PSD biochemical fractions. Figure 1l: pulldown of anti-DCLK in adult brain yields PSD95 (DLG4). |
