Annotation description:"Immunoreactivity was observed in both the presynaptic and postsynaptic areas of excitatory synapses in the cerebellar granular layer and the molecular layer (Figure 8a-d). Mitochondria were also immuno-positive for SENP5 in this region (Figure 8b). Similar distribution profiles, that is, synaptic and mitochondrial localization of SENP5, were observed in the cortex and hippocampus (data not shown).

Immunogold silver-enhanced immunoelectron microscopy indicated that immunogold particles associated with SENP5 were distributed in both pre- and post-synapses, as well as in mitochondria (Figure 9a-d). The proximity to the plasma membrane varied for each particle in the pre- and post-synaptic regions, suggesting that SENP5 associates with a variety of SUMOylated synaptic proteins. These synaptic localization patterns were observed in excitatory synapses and rarely observed in inhibitory synapses (Figure 9c,e,f).

We also found particles located on the outer membranes of mitochondria (Figure 9b,d). This finding was consistent with the observed co-localization of SENP5 and TOM20 in immunocytochemical analyses (Figure 7d). Taken together, our observations suggest that SENP5 plays a role in the regulation of synaptic transmission and mitochondrial function."
