Annotation description: | Figure 3c: co-immunoprecipitation showed NSG2 can interact with AMPAR subunits GluA1 (GRIA1) and GluA2 (GRIA2) in whole-brain lysate of P0 mouse. Figure 4b,c, 5a: whole-cell patch clamp recordings from primary hippocampal neurons with CRISPR knockout of NSG2 showed a decrease in mEPSC frequency (Fig. 4). In contrast, primary hippocampal neurons overexpressing mCherry (control) and NSG2-mCherry showed an increase in mEPSC amplitude (Fig. 5). Figure 4f, 5c, 5e: immunofluorescence confocal imaging of primary hippocampal neurons with CRISPR control and knockout of NSG2 showed a significant reduction in PSD95 (DLG4) fluorescence intensity in neurons expressing NSG2 KO, no change for GluA1 (GRIA1) and some reduction of GluA2 (GRIA2) intensity (Fig 4.). In contrast, primary hippocampal neurons overexpressing mCherry (control) and NSG2-mCherry showed a significant increase in fluorescence intensity for both GluA1 and GluA2 (Fig. 5). Authors conclude: "These data suggest that NSG2 is sufficient to drive increased surface expression of AMPARs into existing functional synapses, and potentially to a small proportion of previously silent synapses." |
