Annotated paper:Bal M, et al. "Reelin mobilizes a VAMP7-dependent synaptic vesicle pool and selectively augments spontaneous neurotransmission" Neuron. 2013 Nov 20;80(4):934-46 PMID:24210904
Figure(s):Figure 1-7
Annotation description:Reelin (RELN) increases spontaneous neurotransmitter release by selectively mobilizing VAMP7 containing synaptic vesicles (and leaves vesicles harboring other vesicular SNAREs relatively unaffected).

Reelin increases the frequency of spontaneous neurotransmission (Fig. 1), selectively augments spontaneous SV trafficking without altering the properties of evoked neurotransmission (Fig. 2) and Reelin-induced increase in spontaneous release requires SNAP-25 but not syb2 (Fig. 4).

Figure 6: Dual color analysis of vesicular SNARE trafficking reveals that VAMP7 specifically responds to Reelin application. "Reelin could selectively increase the rate of trafficking of VAMP7-pHluorin-containing vesicles, while syb2-mOrange labeled vesicles in the same boutons were unaffected (Figure 6E and F). Collectively, these data demonstrate that within a given presynaptic terminal, Reelin selectively mobilizes a subset of VAMP7-containing SVs and leaves vesicles harboring other vesicular SNAREs relatively unaffected."

Figure 7: VAMP7 knockdown is necessary and sufficient to abolish the effect of Reelin.
"To further examine whether VAMP7 expression is indeed required for the effect of Reelin on spontaneous neurotransmitter release, we infected neurons with shRNA constructs directed against VAMP7, VAMP4 or vti1a"
"Uninfected control neurons and neurons infected with the vti1a- and VAMP4-KD constructs still responded to Reelin (Figure 7C-F, I), while knockdown of VAMP7 using either the VAMP7-KD3 or VAMP7-KD4 construct abolished the effect of Reelin"
"Taken together, these results provide further support that Reelin mobilizes a specific subset of SVs containing VAMP7 to facilitate spontaneous neurotransmission."
