Annotation description: | Figure 1a, S1c-d: IgSF21 has two extracellular immunoglobulin-like domains (Ig1 and Ig2) and a putative C-terminal signal sequence for a glycosylphosphatidylinositol (GPI)-anchor modification but no transmembrane domain. Figure S1e-f: IgSF21 is expressed on the cell surface but can be stripped from the surface using phosphatidylinositol-specific phospholipase C (PI-PLC), confirming that IgSF21 is a GPI-anchored protein. Figure 2d: in mouse brain at four postnatal weeks, Igsf21 is preferentially expressed in the deep layers of the cortex, the pyramidal cell layer of the CA1 and CA3 regions of the hippocampus, thalamus and the pons. Figure 2e: At the cellular level, IgSF21 is expressed in neurons but not in glia. Figure 2f: in biochemical subfractions of mouse brain tissue, IgSF21 is enriched in PSD subfractions (similar to PSD95/DLG4). Figure 3: IgSF21 interacts with HA-tagged neurexin2α but not any other neurexin-alpha/beta proteins in COS-7 cells. "IgSF21-Fc-coated beads induced strong accumulation of HA-NRX2α in contacting transfected axons, whereas Fc-coated beads (a negative control) did not (Fig. 3h, i). Together, these data indicate that IgSF21 binds and recruits axonal NRX2α in a trans-interaction manner and that this interaction is essential for the synaptogenic activity of IgSF21." |
