Annotation description: | Figure 3: Defective presynaptic maturation and function in Lnx1-null mice. EM imaging of mossy-fiber synapses in wildtype and Lnx1 -/- mice showed deficits in PSD number per spine, docked synaptic vesicles per release site and synaptic vesicle density. Figure 4: Reverse IPs with knockout and mutated-domain controls showed Lnx1 and EphB1/2 can interact. Figure 5: Lnx1 stabilizes EphB receptors through specific binding sites to prevent their degradation in proteasome. Figure 8: The diminished axon terminals in Lnx1-/- mice (Figure 3, 8a) were restored in mice with both Lnx1-/- and EphB2F620D/F620D mutations. The latter is a constitutively active form of EphB tyrosine kinase. Taken together, authors showed that Lnx1 mediated EphB1 and EphB2 kinase activation promotes mossy fiber presynaptic terminal maturation (Figure 8H). |
