Annotation description: | Upon ARF6 knockdown, synaptic vesicles are depleted and endosome-like cisternae accumulate (Fig1). This can be reverted by expressing shRNA resistant ARF6 (Fig S3) or phenocopied by acute chemical ARF6GEF inhibition by SecinH3 (Fig 3), indication that this is really attributed to impaired AFR6 function. Blocking synaptic activity by TTX also reverts the synaptic vesicle and cisternae number, showing that synaptic activity is needed for the ARF6-dependent SV pool maintenance (Fig4). ARF6 could regulate SV endocytosis and recycling either directly at the plasma membrane and/or from the endosome-like cisternae. |
