Annotation description: | Figure 6d,g: confocal imaging and double immunostaining showed dystrobrevin beta (DTNB) and Gephyrin (GPHN), an inhibitory postsynapse marker, co-localized in the hippocampal dentate gyrus of mouse brain sections. Authors note that "Dtnb was co-labelled with Gephyrin (Fig. 6g) but not with PSD95 (Fig. 6h), indicating that Dtnb was exclusively present at inhibitory postsynapses." Furthermore, DTNB was identified as a binding partner of Dp71 (short product of Dystrophin, DMD, which is also shown to be present in inhibitory synapses in this paper) by IP and both proteins seemed to co-localize at dendritic puncta (Fig. 6). The gephyrin staining had cross reactivity with the nucleus in Figures 3 and 4, on which authors noted: "Arrows indicate strong cross-reactions against nuclear component revealed by anti-Gephyrin antibody which should be ignored". This brings the specificity of the gephyrin antibody into question. Additional annotations of independent experimental evidence would be helpful to support these observations. |
