Annotation description: | Dp71 is a short product of Dystrophin (gene symbol DMD). Figure 3 and 5: HA-Dp71 colocalises with Gephyrin (GPHN), an inhibitory postsynapse marker, in the hippocampal dentate gyrus (DG) granular layer of the transgenic (TG) mouse brain section (Fig. 3) and cultured hippocampal neurons (Fig. 5). Authors write: "Taken together, HA-Dp71 was exclusively localized to inhibitory postsynapses in the cultured hippocampal neurons." WT vs HA-Dp71 control for HA-antibody specificity in Figure 2. Supplementary Figure 6a: "In agreement with all above results, HA-Dp71 puncta were adjacent to GABAergic presynaptic terminals labelled by GAD67" The gephyrin staining had cross reactivity with the nucleus in Figures 3 and 4, on which authors noted: "Arrows indicate strong cross-reactions against nuclear component revealed by anti-Gephyrin antibody which should be ignored". This brings the specificity of the gephyrin antibody into question. However, taken together with the presented co-localization with VGAT (Fig. 5c) and GAD67 (Fig. S6a), localization of Dp71 (DMD) to the postsynapse is likely. |
