Annotation description: | TRAF6 mediates dendritic protrusion formation via neuroplastin (Fig. 5). Knockdown of TRAF6 via siRNA reduces number of formed synapses (panel D). Similarly, treatment with the TRAF6 inhibitor SMI 6860766 reduces the number of dendritic protrusions and Shank2 clusters per protrusion (H-N). Note, TRAF6 effects synapse formation only if applied before day in vitro 9 and not later (Fig. 4). TRAF6 blockage during neuronal development reduces synapse formation affecting neuronal activity (Fig. 6); treatment with SMI TRAF6 reduces the number of excitatory synapses and there is reduced network activity in SMI TRAF6-treated hippocampal neurons grown on multi-electrode arrays. |
