| Annotation description: | Traf6 is the E3 ubiquitin ligase that mediates constitutive K63 ubiquitination of PSD-95 (DLG4). Together with its deubiquitinase counterpart CYLD (SynGO annotation #4535) it controls PSD-95's clustering efficacy in spines and in turn controls cell surface availability of NMDA receptors in the postsynaptic membrane. Fig. 5D,E shows reduction of PSD-95 clustering upon Traf6 knock-down. Further: "Knocking down TRAF6 prevented the AP5-elicited increase of PSD-95 ubiquitination (Fig. 7G), supporting the notion that TRAF6 mediates constitutive K63 ubiquitination of PSD-95." |
