Annotation description:"Immunofluorescence staining also revealed a wide distribution of Sarm1 protein (Fig. 3, C and F) with a punctate pattern in soma and dendrites. Compared with PSD-95, a postsynaptic density marker, the Sarm1 puncta were smaller in size and widely distributed along dendrites (Fig. 3 F, left). However, some Sarm1 puncta colocalized with PSD-95 (Fig. 3 F, top right, arrowheads), whereas others were surrounded by or adjacent to PSD-95 (Fig. 3 F, top right, arrows).

The correlation coefficient of PSD-95 and Sarm1 colocalization was 0.26. Although the colocalization coefficient was not high, shifting the image overlap by 1 or 1.66 μm significantly reduced the colocalization coefficient to 0.18 or 0.15, respectively (Fig. 3 F, bottom right; original vs. 1-μm shift, P = 0.0096; original vs. 1.66-μm shift, P = 0.0012).

These analyses suggest that Sarm1 distributes along dendrites and that a fraction of Sarm1 is present at synapses." (text p. 773).

In addition, the paper shows the presence of Sarm1 in lysed synaptosomal membranes of mouse brain (Fig. 3E), in which PSD-95 is highly enriched. The data do not allow final conclusions on whether Salm1 is presynaptic or postsynaptic or both. Actually, the enzyme is widely distributed in general in the cell.
