Annotation description: | Figure 1: whole-cell voltage-clamp recordings of pyramidal neurons in hippocampal slices show diminished presynaptic release in BAD, BAX and caspase-3 (CASP3) KO mice, by demonstrating that (A) mEPSC frequency is reduced, (B) Paired-pulse ratio is increased and (C) EPSC amplitude is reduced in the different KO models compared to WT. Figure 2: whole cell recordings and electron microscopy were performed on hippocampal slices from BAD, BAX and caspase-3 KO mice. Analysis of sucrose evoked responses with electrophysiology, and the synaptic vesicle pool with EM shows evidence for a reduced readily releasable pool in the BAD, BAX and caspase-3 KO mice. Furthermore, authors show evidence for autophagy controlling the size of synaptic vesicle pools in presynaptic neurons (Figure 3, 5, 6) as well as evidence for activity-dependent regulation of autophagy by caspase-3, influencing presynaptic release following high-frequency stimulation (Figure 9). Given the role of caspase-3 in regulating the synaptic vesicle pool, here shown to be likely through the regulation of apoptosis through the BAD-BAX-caspase-3 cascade, the ontology term "regulation of synaptic vesicle cycle" was selected. |
