Annotation description:Figure 1: A calpain-sensitive marker, pYSCS, was designed with an intervening sequence from the μ-calpain (CAPN1, calpain 1) cleavage site in α-spectrin. A PDZ cognate sequence that binds to PSD95 was added at the 3' end to facilitate retention of the expressed protein at postsynaptic sites.

Figure 4c: Expression of YSCS in dissociated hippocampal neurons showed YSCS is concentrated in spine heads.

Figure 5: "The utility of pYSCS as a reporter of glutamate receptor-induced calpain activation was initially examined in real-time imaging experiments in cultures of dissociated hippocampal neurons. Images collected before and 3 to 5 min after exposure of neurons to glutamate or NMDA revealed a substantial decrease in the FRET ratio in dendritic spines (Fig. 5 A-F)"
