Annotation description:This study provides extensive characterization of PLC beta1 localization in brain, using immuno-electron microscopy (HRP labeling and immunogold pre- and postembedding).

In CA1 pyramidal cells, PLC beta1 was predominantly detected in somatodendritic compartments, including dendritic spines (Figure 3, 4). No signal was observed in presynaptic terminals. Note that most signal is cytoplasmatic, although some membrane staining is observed. The latter might be due to activity dependent recruitment of PLC to the plasma membrane via its PH or C2 domains. In the cerebellum, PLC beta1 appears to be largely absent from the synapse (figure 8).

A subcellular fractionation for PLC beta1 is also provided (Figure 3A). This result is in agreement with the EM data, but lacks controls.

Extensive characterization of the antibody is provided, including knockout controls in western blot (Figure 1A, immunofluorescence (Figure 1B) and immuno-EM (Figure 3,4).
