Annotation description: | Figure 1,S1: Endogenous mGluR2 (GRM2) was tagged with a pH-sensitive variant of GFP using a CRISPR/Cas9-mediated knock-in approach. Super-resolution imaging of hippocampal neuronal cultures showed mGluR2 was localized both in axons and dendrites (Fig. S1A), and detected both in the axon shaft and within synaptic boutons (Fig. 1A). Co-staining with Bassoon (BSN) showed mGluR2 was localized near but not at the active zone (Fig. 1A). Figure 2,S2: Fluorescence recovery after photobleaching (FRAP) experiments showed surface-expressed mGluR2 was highly mobile and diffusely spread throughout the axon. A model for mGluRs 2 and 7 is proposed in Figure 6. Based on the known transmembrane topology of the protein, the "integral component of ... membrane" term was chosen. |
