Annotation description:Calmodulin (CaM) immunostaining in P9 WT mice showed strong overlap with VGLUT1 at calyx of Held synapses, which was significantly reduced after Calm2 knockout (Figure 6B).

Not all Calmodulin expression was abolished after knockout of Calm2. Because Calmodulin 1, 2 and 3 have 100% sequence similarity, the Calmodulin antibody targets all 3 CaM genes and can stain remaining Calm1/3 expressed at the synapse after Calm2 knockout.

This conclusion is supported by Figure 7A and 7B which identify marked reduction in calmodulin immunoreactivity in hippocampal neurons in culture in the CaM2 knockout likely due to Cam1 and CaM3 expression.
