Annotation description: | Figure 1C: Syt3 is predominantly in dendrites in hippocampal neurons transfected with GFP and immunostained with MAP2 to mark dendrites (MAP2-positive) or axons (GFP-positive/MAP2-negative). Figure 1D: Syt3 localizes to synapses marked by synaptophysin and PSD95 (top) or GluA1 (bottom) in hippocampal cultures. Figure S1I-J: In subcellular fractions, Syt3 is associated with synaptosomal plasma membranes and not with synaptic vesicles. Immuno-organelle isolation of synaptic vesicles with antibodies to Syt1 or Syb2 further confirmed that Syt3 is not present on synaptic vesicles. Figure 1E-F: Synaptosomes form in such a way that the presynaptic terminal seals, trapping synaptic vesicles and other presynaptic components inside, whereas the postsynaptic membrane does not reseal. Trypsin cleavage assay of synaptosomes (PMID: 23622064) can be used to deduce whether exposure to trypsin cleaves the protein of interest based on its localization in either the presynapse or postsynapse. The current experiment shows that postsynaptic proteins, including Syt3, are cleaved upon trypsin treatment whereas presynaptic proteins are protected indicative of a postsynaptic localization of SYT3. The antibody against SYT3 was verified in a SYT3 KO mice (Figure 1A). |
