Annotation description:CPEB3 is strongly enriched in the PSD fraction of mouse brain. In addition, the protein is detected by immunofluorescence in DIV21 hippocampal neurons, where it is juxtaposed to synaptophysin (Fig. 5A), which is taken to indicate a pre-synaptic localisation. Since, however, the optical resolution in the immunofluorescence is not sufficient to decide if the in-vitro staining is pre- or post-synaptic, there seems to be more evidence for a post-synaptic localisation.

This annotation is also supported by Pavlopoulos et al., Cell 2011.

- Literal: "Finally, both CPEB3 and CPEB4, when fused to GFP, were detected in dendrites, often as puncta (Figure 5C)"
- Literal: "Taken together, these data show that CPEB3 and CPEB4 are expressed in only partially overlapping regions of the brain; within hippocampal neurons, where both are expressed, they appear to be synaptic."
- The mRNA translation data is too poorly shown in this paper lacking clear synaptic relevance although looking at AMPAR translation.

Additional literature:Dendritic, possibly post-synaptic localisation in mouse CA1 by immunofluorescence: Fig. 5B. The protein is clearly dendritic and forms puncta that are next to but distinct from synaptophysin puncta => likely post-synaptic sites. @ PMID:22153079

CPEB3 is detected in snyptosomes: local translation is dis-inhibited by NMDA-induced calpain degradation. Note, however, that the synaptosomes here are not highly purified, this fraction very likely contains all microsomes.
Rat brain was used, no further specification of strain , sex, or age is given. @ PMID:22711986
