Annotation description: | By using conditional Lphn3 KO mice, the authors sparsely express Cre to compare Lphn3 KO primary neurons to adjacent wild type neurons in culture. Loss of Lphn3 leads to a reduction in excitatory postsynaptic density in excitatory synapses (Fig 1D) and not in inhibitory synapses (Fig 1E). This can be rescued by re-expressing Lphn3 but not by re-expressing mutants that block binding of FLRT (4A mutation) or teneurin (ΔLEC mutation) (Fig 4C), known presynaptic partners of Lphn3. This results are confirmed in intact tissue by analyzing spine density in hippocampal neurons sparsely transduced with Cre-GFP virus and filled with biocytin. This reveals that Lphn3 only controls spine density in 2 of the 3 major synaptic hippocampal layers (S. oriens and S. radiatum but not in S. lacunosum-moleculare). In addition, to demonstrate that Lphn3 is a potent inducer of presynaptic excitatory specializations into regions expressing Lphn3, authors use cocultured neurons and HEK cells in an artificial synapse formation assay. HEK cells expressing GFP and Lphn3 show strong vGlut staining, indicating that presynaptic terminals of cultured neurons can synapse into HEK cells that express Lphn3. Control with only GFP expression does not induce any vGlut staining at the HEK cell surface (Fig S9A, S9C). |
