Annotation description: | Loss of RBP1,2 in double knockout mice show impaired presynaptic localization of BKα-channel protein (Figure 8). Presynaptic BK channels act as negative regulators of neurotransmitter release by hyperpolarizing the plasma membrane in response to AP-stimulated depolarization and Ca2+ entry through CaV channels. They show also that BK currents (amplitude and density) are abolished in the RBP1,2 double knockout mice (Figure 7). Therefore, RIM-binding proteins 1 & 2 control potassium currents and membrane potential presynaptically. 18/01/2021 Dnyanada: BK are large-conductance calcium-activated potassium channels that are enriched in the presynapse (Robitaille et al, 1993a; Knaus et al, 1996; Zhou et al, 1999; Hu et al, 2001; Ishikawa et al, 2003; Misonou et al, 2006; Nakamura & Takahashi, 2007). The current paper shows that deletion of RBP decreases and decelerates presynaptic BK currents (Figure 5, 6) by impairing presynaptic localisation of BK channels (Figure 8; as described in the annotation). Based on this, the ontology term "regulation of presynaptic membrane potential" was chosen. Note: FigS2C uses IP to demonstrate that RIMBP1,2 dKO does not alter the levels of BK channels as compared to WT. However, it is interesting to note that RIMBP1,2dKO samples stained against anti-RIMBP2 show a light band which means that RIMBP1,2 dKO model might not be completely abrogating the expression of RIMBP2. |
