Annotated paper: | Huang HS, et al. "Snx14 regulates neuronal excitability, promotes synaptic transmission, and is imprinted in the brain of mice" PLoS One. 2014 May 23;9(5):e98383 PMID:24859318 |
Figure(s): | Fig 3 and 4 |
Annotation description: | Fig3A-B: The effectiveness of lentiviral Snx14 shRNA knockdown was verified by immunocytochemistry analysis followed by puromycin treatment (inhibitor of protein translation). As expected, neurons transfected with lentiviral Snx14 shRNA showed reduced signal when stained with anti-Snx14. There was no difference in anti-snx14 expression pattern in neurons transfected with vehicle or scrambled snx14 shRNA. Fig3D: Snx14 knockdown reduces intrinsic excitability (~60%) of pyramidal neurons. Fig4A-B: mEPSC and mIPSC recordings following Snx14 knockdown showed a reduction in mEPSC and mIPSC frequency, but not amplitude. The paper does not further investigate the mechanism by which Snx14 KD affects mEPSC and mIPSC data. Hence, the general term "postsynaptic modulation of chemical synaptic transmission" was chosen. |
