Annotation description: | Fig 3: Subcellular fractions were obtained from adult rat cerebral cortex and the distribution of FAM81A protein was monitored using two antibodies (top two panels). FAM81A immunopositive band at ~42 kDa was found to be highly enriched in the PSD fraction, in parallel with PSD-95, whereas the nuclear fraction showed very little labelling at that position with either Ab1 or Ab2. Other bands in the nuclear fraction that were immunopositive with both Ab1 and Ab2 were further analyzed by mass spectrometry, but no peptides corresponding to FAM81A were identified (data not shown). Fig 4: Immuno-electron microscopy on excitatory synapses from adult mice hippocampus (Fig 4A-C) and cerebral cortex (Fig D-E) showed specific labelling of FAM81A protein in the postsynaptic density. Fig 5: Laminar distribution of FAM81A label was concentrated at the cytoplasmic edge of the PSD core, with a mean distance of 34.1±1.1 nm from the postsynaptic membrane in the hippocampus and 32.6±1.1 nm from the postsynaptic membrane in the cerebral cortex. Supplementary Fig 3: Immunoreaction distribution profile in laminar layers of PSD of FAM81A was compared with that of PSD95, GKAP and Homer. FAM81A labelling follows the same pattern of distribution as GKAP, while PSD95 and Homer show distribution in the PSD core and PSD pallium. Fig4, Fig5 and Fig S3, the authors claim that FAM81A is majorly concentrated at the cytoplasmic core of PSD. Uniprot does not show any transmembrane domains for this protein (nor for its perhaps better annotated human ortholog). Based on this, the SynGO term "postsynaptic density, intracellular component" was used. The authors do not comment on the specificity of the FAM81A antibody. |
