Annotation description:Authors show in dissociated hippocampal cultures that AKAP150 plays a crucial role in mediating NMDA-induced AMPA receptor endocytosis. Interaction of AKAP150 and PSD95 is crucial for mediating this process, as a point mutation in PSD95 that prevents binding to AKAP150 is sufficient to prevent the endocytosis of AMPA triggered by NMDA treatment. Moreover, over expression of AKAP150 lacking the binding region to calcineurin, a protein known to be involved in NMDAR-dependent LTD and AMPA receptor endocytosis, also completely prevents the AMPA receptor endocytosis. These results suggest that AKAP150 plays a crucial role in the process by positioning calcineurin in the vicinity of NMDAR by interacting with PSD95.

- The postulated role of AKAP150 in AMPAR-endocytosis is "Our results suggest that PSD-95's interaction with AKAP150 is critical for NMDAR-triggered AMPAR endocytosis and LTD, possibly because these scaffolds position calcineurin in the appropriate subsynaptic domain." Since AKAP150 is presented as a critical trigger I propose to check the "regulation of...." box.
