Annotation description:Figure 1: Electron micrographs showing RTN4 immunoreactivity in the spinal cord motoneurons of adult rats, detected with pre-embedding immunogold-silver cytochemistry (A-C) and post-embedding immunogold cytochemistry (D). Immunogold particles indicating RTN4 immunoreactivity could be found in dendritic profiles (Den) and in close apposition to the postsynaptic density (thin arrows in A-C) and the postsynaptic dense bodies (thin arrows in C and D) at both symmetric (thick arrow in A and B) and asymmetric synapses (thick arrow in C and D).

Note on the antibody used: Antibody was generated against an RTN4 specific C-terminus 572-amino acid fragment (621-1192). The antibody is further described in PMID: 12577319.

Figure 1: Western blot of the lysate of the mock plasmid transfected COS-7L cells showed negative staining with the RTN4 antibody (lane 1 of Fig. 1A). That of the COS-7L cells with full-length hnogo-A (human RTN4) transfection displayed several bands, with the topmost band at 220 kDa, which matched the size of Nogo-A (lane 2 of Fig. 1A). Western blot of the spinal cord showed a very strong band at 220 kDa and a faint band at the lower end (lane 3 of Fig. 1A).
