-In Fig 1, different electrophysiological recordings are carried out to measure Ca2+-activated Cl conductance in motoneurons. In established conditions to prevent Na+ and K+ currents, we can observe an outward current (depolarization) and tail inward (repolarization) after applying voltage ramps from -100 mV to +20 mV. They developed these assays in motoneurons electroporated with control siRNA or specific siRNAs against Tmem16 genes, observing a significant decrease in outward and inward currents for the siRNA-Tmem16f condition and not for the others (Figs. D and E), which suggest the specific role of TMEM16F for Cl conductance in motoneurons.

Notes:

-In Fig. 2B, they showed successful siRNAs electroporation observing rhodamine positive motoneurons before to carry out the electrophysiological recordings.

- Moreover, we observed a similar result to Tmem16f RNA inhibition in (Figs. 1F-1I) when they use for the experiments motoneurons with targeted deletion of exon 3 of Tmem16 (strategy in Fig. S2A)

11/01/2021 Dnyanada: Members of Tmem16/Anoctamin family (calcium-activated chloride channels; CACCs) lie in close proximity of calcium channels and regulate the activity of chloride channels (PMID: 22500639). The paper demonstrates that TMEM16F regulates the functional expression of CACCs in motorneurons and as a product governs Ca2+ activated Cl conductance in motorneurons. The paper also shows that TMEM16F might be localised in the postsynapse. Based on the role of TMEM16F in chloride conductance and its localisation in the postsynapse, the term "regulation of postsynaptic membrane potential" was chosen.
