Annotation description: | "TMEM16F / Anoctamin-6 could be a post-synaptic component of C-boutons" Several experiments in Fig 2 show that TMEM16F protein is an alfa-motoneuron marker, which faces cholinergic C-boutons synapses. - "By staining TMEM16F was observed in choline acetyltransferase-positive (ChAT+) motoneurons, specifically located in the ventral horn laminae VIII and IX. This protein can be observed forming clusters at the plasma membrane of soma and dendrites (Figs. 2A and B)" - "Co-staining of TMEM16F with a plasma membrane marker of motoneurons (Na+/K+-ATPase alfa1) is shown in (Fig. 2F) verifying its localization. In Fig 2G, TMEM16F protein is observed on the opposite side to the presynaptic marker Synaptophysin, which could indicate a postsynaptic localization, although strong evidence would be necessary. Moreover, TMEM16F clusters are found facing the vesicular acetylcholine transporter (VAChT) pre-synaptic clusters (Fig. 2I), being VAChT a marker of synaptic cholinergic C-boutons. |
