Annotation description: | Figure 7, A and B: Fluorescence microscopy targeted towards intracellular and total GluA2 was used to calculate the GluA2 internalisation index. TSPAN7 Knockdown demonstrated an increase in the GluA2 internalisation index indicating its role as a regulator in GluA2 trafficking in the post-synapse. To ascertain that the effect was a true representation of endocytosis of GluA2 receptor, a similar experiment in the presence of dynamin inhibitor Dynasore was carried out. Dynasore abolished the differences in the internalization index between TSPAN7-knockdown and scrambled siRNA14 neurons. Figure 8 A: Rescue WT rescues the effect of TSPAN7 knockdown by reducing the internalization index close to control levels. Overexpression of TSPAN7 reduces the GluA2 internalization index compared to control. |
