Annotation description: | C. elegans RAP-2 GTPase (rap-2) is involved in regulating synapse formation. RAP-2 null mutants and gain-of-function mutants (constitutive GTP-form of rap2) exhibit synaptic tiling defects indicating that the cycling between GTP- and GDP-forms is required for the regulation of spatial patterning of synapses (Fig. 1). Expression of rap-2 in the postsynaptic muscle cells cannot rescue the tiling defect, whereas presynaptic expression in DA neurons can rescue the phenotype (Fig. 2). Fluorescence Lifetime Imaging Microscopy (FLIM)-based FRET (Förster Resonance Energy Transfer) measurements using human EGFP-Rap2A were performed to test the localization of Rap-2 activity with the help of a construct (mRFP-RaIGDS(RBD)-mRFP) that specifically binds to GFP-Rap2, but not GDP-Rap2 (Assay explained in Fig. 4A). Human Rap2 had higher activity in synaptic regions compared to non-synaptic domains (Fig. 4). 19/01/2021 Dnyanada: As Human Rap2a cDNA was used for generating RAP2A mutant c.elegans lines, P10114 (RAP2A_HUMAN) was chosen. |
