Annotation description: | De Jong et. al demonstrate that RIM1 binds to the plasma membrane of the presynaptic active zone. - In STED microscopy, RIM colocalizes with Bassoon, a well established active zone marker. This colocalization is true for both endogenous RIM as well as RIM1 rescue constructs, which will be used in the remainder of the study (Figure 1 G-M) - The C2B domain binds to PIP2, a phospholipid that is present in the plasma membrane. The interaction is demonstrated with NMR spectroscopy, lipid cosedimentation and lipid cofloatation assays (Figure 3A-F). Additionally, a mutation is identified that abolishes PIP2 binding (Figure 4). - The PIP2-binding mutation in the C2B domain inhibits the function of RIM1 C2B in action-potential induced synaptic transmission (Figure 5). |
