Annotation description:Sclip et al provide multiple lines of evidence to demonstrate that RIMBP2 regulates BK-channels at the presynapse:
- Using yeast-2-hybrid and immunoprecipitation it is shown that RIMBP2 interacts with BK-alpha. (Figure 1)
-In HEK cells, RIMBP2 recruits BK-alpha to the plasma membrane (figure 1 D, E) and increases the BK-alpha mediated K+ current at lower membrane voltages (figure 4).
-In line with the observations above, knockout of RIMBP1 and RIMBP2 (DKO) reduces the BK-mediated K+ current in the Calyx of Held (Figure 6A-H), and slow down the activation kinetics of these currents (Figure 6J-M).
-The DKO also reduces the recruitment of BK-alpha to the presynaptic terminal (Figure 8).
