Annotation description: | Developed an antibody against the pore forming region of Cav1.2 that was used in light and electron microscopy of fixed and stained rat brain sections. Immunoperoxidase labeling for the Cav1.2 antibody was clustered in patches in the cytoplasm of dendritic shafts and along the plasma membrane of dendrites (Fig. 3A-F). Labeled dendrites often formed synaptic contacts with unlabeled axon terminals, and most of these synapses were asymmetric (Fig. 3A,D,E). Cav1.2 was observed less frequently in dendritic spines where the peroxidase reaction product was observed mostly in the cytoplasm or regions of the plasma membrane away from the postsynaptic density (Fig. 4A-D). The pre-embedding immunogold method was used to localize Cav1.2 more discretely. Immunogold particles were mostly localized in the cytoplasm of dendrites but a significant portion of the labeling was also associated with the plasma membrane (Fig.7A,B), particularly in small dendrites. In dendritic spines of CA1 and CA3 neurons immunogoldparticles were predominantly intracellular (Table 2). 21/6/2017 Pim - I regard the predominant intracellular localization of Cav21 in these preparation as reflecting a non-canonical localization for this protein (protein synthesis, inactive pool) and I refrain from annotation of this localization. |
