Annotation description: | Analyzing parallel fiber inputs to Purkinje cells, Fig.3A shows that most vGLUT1 puncta overlapped with the punctate contactin-6 staining. Dense contactin-6 puncta were apposed to dendritic mGluR1a of Purkinje cells. These data support a presynaptic localization of contactin-6. 21/6/2017 Pim - The rationale is: (1) protein expressed in axons (parallel fibers). (2) colocalization with pre- and postsynaptic marker proteins. (3) localization in synaptosomal preparations. - Although superresolution data is not presented, based on cell type expression and colocalization data the authors conclude: "Col-lectively, these results indicate that NB-3 is localized presynaptically at glutamatergic synapses." |

Additional literature: | Hydrophobicity analysis (Kyte and Doolittle, 1982) revealed that hNB-3 has the typical features observed in GPI-anchored membrane proteins @ PMID:9486763 The authors show that endogenous NB-3 protein in cultured cortical neurons from wild-type is expressed at the surface, a disrupted GPI consensus site in NB-3 strongly reduces membrane expression in COS cells. The authors do show that PTPalpha is able to stabilize NB-3 at the plasma membrane independent of the GPI link. @ PMID:21622556 |
