Annotation description: | Primary hippocampal neurons were fixed and stained with mouse monoclonal anti-beta1 antibody. anti-beta1 staining could be detected in the soma and dendrites (Fig. 1B). The staining appeared punctate along dendrites (Fig. 1C) and showed partial co-localization with antibody staining for a synaptic marker (synapsin) although the majority of beta1 clusters appeared to be adjacent to synapsin clusters. Object-based colocalization analysis of 24 DIV neurons was also used to support partial overlap of beta1 subunits with synapsin (% of β clusters colocalized with synapsin ± 95% confidence intervals (n): β1, 46 ± 4 (21). Although beta 1 expression in axons is lower than some of the other subunits it can be detected. This data shows that some beta1 is axonal and presynaptic but this is a minority of the protein. Expression of tagged beta1 that provides better resolution shows clear overalap with presynaptic markers (Fig 4) Supplementary Fig 1. Specificity of anti-beta antibodies tested. Dysgenic myotubes were transfected with a V5 epitope containing version of the beta1 subunit. Cells were fixed and stained with antibodies against all 4 subunits but the antibodies did not label non-transfected neighboring cells, or cross-react with cells expressing any of the other beta isoforms (Suppl.
