Annotation description: | Preembedding immunogold electron microscopy (EM) on lamina I-III of L4-L6 spinal cord sections was used to study the subcellular localization of alpha_2 delta-1. Alpha_2 delta-1 was observed at the plasma membrane of presynaptic nerve terminals of DRG neurons, at the plasma membrane of postsynaptic dorsal horn neurons and also at intracellular dendritic sites (Fig 1E, F). 27/4/2017 Pim - In discussion with the expert we have establised that PMID:20080692 (Fig.4G) shows that the Ca-channel alpha_2 delta-1 subunit may have a TM domain predicted, but that A GPI anchor provides most likely the link to the plasma membrane. Additional literature: | Paraffin-embedded sections of rat brain were stained with primary antibody specific for alpha_2 delta-1 and a horseradish peroxidase coupled secondary. The alpha_2 delta-1 protein was observed at the neuropil, and at a much lower level in somata (Fig 6). @ PMID:18616987 Provides evidence that alpha_2 delta-1 is GPI anchored in the plasma membrane of cells. alpha_2 delta-1 is expressed on the plasma membrane of DRGs in culture (Fig. 4E, immunofluorescence) and incubation with phosphatidylinositol-phospholipase C (PI-PLC) causes it to be cleaved from the surface of cells.
