Annotation description: | The authors reported that VAMP4 is needed for Activity-dependent Bulk endocytosis (ADBE), a pathway involved in the recovery of synaptic vesicles after intense stimulation. To determine whether VAMP4 is involved in bulk endocytosis, the authors inhibited GSK3, a kinase known to be active in ADBE, and found that inhibition of ADBE greatly diminishes the proportion of nerve terminals displaying activity-dependent slow VAMP4-pHluorin downstrokes, suggesting this downstroke reflects bulk endosome acidification. It was shown that Vamp4 knockdown in neuron abolishes the TMR-dextran uptake and this can be fully recovered by expression of wild-type Vamp4, documenting that Vamp4 is involved in Activity dependent Bulk endovytosis (ADBE). Also, it was shown that interaction vamp4 to endocytosis adaptor proteins is necessary for its function in ADBE (Fig. 7) and mutation of the binding site of vamp4 to adaptor proteins decreases the ADBE (Fig.8). |
